Abstract: | Objective: We investigated the association of microRNA-126 (miR-126) with ocular ischemia in rats with chronic bilateral common carotid artery occlusion (BCCAO), and in a cell culture of primary rat retinal capillary endothelial cells (RRCECs). Methods: A total of 60 Brown Norway rats were divided into four groups: BCCAO group; BCCAO group with administration of an intravitreal injection of the lentiviral vector (LV)-pre-miR-126 inhibitor-green fluorescent protein (GFP) in one eye and of LV-miR control-GFP in the other eye; sham surgical group; control group. Results: In the BCCAO group without injection and with LV-miR-control, the levels of miR-126, phosphorylated forms of extracellular-signal regulated kinase (ERK), vascular endothelial growth factor (VEGF), and basic fibroblast growth factor (bFGF) were significantly higher, than those in the BCCAO group with LV-pre-miR-126-inhibitor and the sham surgical group. The application of miR-126 inhibitor induced upregulation of Spred-1 protein levels. Seventy-two hours after RRCECs transfection, miR-126 inhibitor downregulated phosphorylated ERK, VEGF, and bFGF. Conclusions: Experimental chronic ocular ischemia is associated with upregulation of miR-126 and increased phosphorylated forms of ERK, VEGF, and bFGF and a decrease in Spred-1 rates as the target of miR-126. Therefore, miR-126 may play a transcriptional role in the pathogenesis of ischemic retinal neovascularization. |
